NMN (Nicotinamide Mononucleotide): Benefits, Dosage and Safety

NMN is a molecule the body uses to make NAD+, a coenzyme involved in energy metabolism and cell repair whose levels fall with age. Human trials show that NMN supplements reliably raise blood NAD+ levels and appear safe in the short term, but clear benefits for ageing, energy or disease have not yet been shown in people. Most of the exciting results come from animal studies.

What is Nicotinamide Mononucleotide (NMN)?

Nicotinamide mononucleotide (NMN) is a nucleotide made from nicotinamide, a form of vitamin B3. Inside cells it is converted to nicotinamide adenine dinucleotide (NAD+), which hundreds of enzymes need, including those that produce energy in mitochondria and repair DNA.

NAD+ levels decline with age in several tissues. In mice, raising NAD+ with NMN improved several markers of ageing and metabolism, which led to strong interest in NMN as an anti-ageing supplement.

NMN occurs in tiny amounts in foods such as broccoli, cabbage, edamame and avocado. Supplements provide far more, usually 250–1,000 mg per dose. Nicotinamide riboside (NR) is a related precursor that also raises NAD+.

Typical uses

Raising NAD+ levels

Human trials consistently show that NMN at 250–1,000 mg per day increases NAD+ and related metabolites in blood within a few weeks. Whether this translates into health benefits is still unknown.

Healthy ageing and physical function

Small trials in middle-aged and older adults found modest improvements in walking speed, grip strength or endurance in some studies, and no change in others. The trials are short and small.

Metabolic health

One small trial in prediabetic postmenopausal women found improved muscle insulin sensitivity after 10 weeks of 250 mg per day. Other metabolic markers such as weight, blood pressure and cholesterol have generally not changed.

Energy, sleep and skin

NMN is marketed for energy, sleep quality and skin. Evidence in people is preliminary.

How strong is the evidence?

  • StrongRaising blood NAD+. Consistent finding across human trials.
  • LimitedMuscle insulin sensitivity. One small RCT.
  • LimitedPhysical performance in older adults. Small trials with mixed results.
  • InsufficientLifespan or anti-ageing effects in humans. Animal data only.

Typical doses

ContextDoseNotes
Human trials250–1,000 mg per dayMost studies lasted 4–12 weeks.
Safety studiesUp to 1,200 mg per dayWell tolerated in trials of up to about 12 weeks.
  • Many people take NMN in the morning, although timing has not been shown to matter in trials.
  • Sublingual and liposomal products are marketed for better absorption, but there is no good evidence they work better than ordinary capsules.
  • NMN-plus-NR or NAD+ "stack" products contain less NMN per capsule; check the milligrams of each ingredient.

Safety, side effects and interactions

Safety

  • NMN was well tolerated in human trials at up to 1,200 mg per day for up to about 3 months; long-term safety is unknown.
  • Reported side effects are uncommon and mild: nausea, flushing, headache or stomach upset.
  • Raising NAD+ could in theory support the growth of existing tumours, a concern raised from lab research; people with cancer should talk to their oncologist first.
  • Regulatory status: in 2022 the US FDA said NMN could not be sold as a dietary supplement because it was being studied as a drug. In 2025 the FDA reversed this position, and NMN supplements are sold in the US again. Rules differ in other countries.

Interactions

  • No well-documented drug interactions in humans. Ask your pharmacist if you take diabetes medicines, as NMN may affect insulin sensitivity.
  • Combining NMN with high-dose niacin or other NAD+ precursors adds to the total B3-related intake.

Who should avoid it or ask a doctor first

  • Pregnancy and breastfeeding: no safety data.
  • Children and teenagers.
  • People with active cancer, unless their oncologist agrees.

Frequently asked questions

Does NMN reverse ageing?

No human study has shown that NMN slows or reverses ageing. It raises NAD+ levels, and some small trials show modest changes in function, but anti-ageing claims are based mainly on animal research.

NMN or NR: which is better?

Both raise NAD+ levels in people. There are no good head-to-head human trials showing one is better. NR has been studied for longer; NMN is newer to the market.

How long does it take NMN to work?

Blood NAD+ levels rise within 1–2 weeks of daily use. Whether you feel any different is uncertain; many trials found no change in how people felt.

Is NMN legal in the US?

Yes. After saying in 2022 that NMN was excluded from supplements, the FDA reversed that position in 2025, so NMN can again be sold as a dietary supplement.

Should NMN be taken with resveratrol?

The pairing comes from early animal research. There is no good human evidence that adding resveratrol improves NMN results.

Choosing a Nicotinamide Mononucleotide (NMN) supplement

  • Check milligrams of NMN per capsule and the purity claim (for example 99% NMN) backed by a lab certificate.
  • Choose products with third-party testing; independent analyses have found NMN products with far less NMN than labelled.
  • Store away from heat and humidity; NMN can degrade over time.
